Evaluation of Different Molecular Markers for Genotyping Non-Saccharomyces Wine Yeast Species

چکیده
Wine quality is determined by the particular yeast strains prevailing at various stages of fermentation. Therefore, ability to make an easy, fast, and unambiguous discrimination yeasts strain level great importance. Here, tandem repeat-tRNA (TRtRNA) method with 5GAC or ISSR-MB primer sets random amplified polymorphic DNA (RAPD) analysis (GTG)3, R5, RF2 oligonucleotides were tested on non-Saccharomyces wine species. The TRtRNA-PCR employing showed highest capacity in discriminating Lachancea thermotolerans Metschnikowia pulcherrima isolates. RAPD was most efficient resolving Starmerella bacillaris isolates, although it produced few bands. R5 discriminate among Issatchenkia orientalis, Hanseniaspora guilliermondii, Pichia anomala either exhibited Torulaspora delbrueckii (GTG)3 for H. uvarum Here we concluded that both RAPD-PCR offer rapid means typing However, each performs better a given species when paired set. present results can be useful research fast fingerprinting yeasts.

Biogeographical characterization of Saccharomyces cerevisiae wine yeast by molecular methods
Biogeography is the descriptive and explanatory study of spatial patterns and processes involved in the distribution of biodiversity. Without biogeography, it would be difficult to study the diversity of microorganisms because there would be no way to visualize patterns in variation. Saccharomyces cerevisiae, "the wine yeast," is the most important species involved in alcoholic fermentation, an...
متن کاملMolecular typing techniques as a tool to differentiate non-Saccharomyces wine species.
A total of 32 yeast strains belonging to four non-Saccharomyces species associated with winemaking was characterized by different molecular techniques. The PCR amplification of 18S rRNA-coding DNA and nontranscribed spacer, followed by restriction analysis with the endonucleases HaeIII and MspI, and PCR fingerprinting with microsatellite primers (GAC)(5) and (GTG)(5) were used. The methods used...
متن کاملIntroducing a New Breed of Wine Yeast: Interspecific Hybridisation between a Commercial Saccharomyces cerevisiae Wine Yeast and Saccharomyces mikatae
Interspecific hybrids are commonplace in agriculture and horticulture; bread wheat and grapefruit are but two examples. The benefits derived from interspecific hybridisation include the potential of generating advantageous transgressive phenotypes. This paper describes the generation of a new breed of wine yeast by interspecific hybridisation between a commercial Saccharomyces cerevisiae wine y...

Background and Purpose: By using advanced detection/identification methods, the list of emerging uncommon opportunistic yeast infections is rapidly expanding worldwide. Our aim in the present study was sequence-based species delineation of previously unidentified yeasts obtained from a clinically yeast collection. Materials and Methods: A total of twenty three out of the 855 (5.7%) yeast isolat...
متن کاملSelection of non-Saccharomyces yeast strains for reducing alcohol levels in wine by sugar respiration.
Respiration of sugars by non-Saccharomyces yeasts has been recently proposed for lowering alcohol levels in wine. Development of industrial fermentation processes based on such an approach requires, amongst other steps, the identification of yeast strains which are able to grow and respire under the relatively harsh conditions found in grape must. This work describes the characterization of a c...
